I can't recover hard drive space after deleting files

Specs: Windows 7
When I delete a file my hard drive it doesn't decrease space. Instead it keeps telling me my hard drive space is full when I know that's not the truth. I've tried going into safe mode and scanning with Malwarebytes and it didn't pick up anything. Then I tried Superantispyware and it picked up two Browser Hijack Objects and regular cookies. I thought maybe I caught it but when I booted by in regular mode my space still didn't decrease after deleting a huge file. What should I do next?

What is the total capacity of the harddrive and how large is the file you deleted?

Excellent job scanning with both Malwarebytes and SuperAntispyware before posting your question!

The size of the drive is 500 GB. I tried deleting a 1 gb video file and the extra space still didn't become available. I noticed that now I have 1.37 GB's of space left when I know I should have at least have 200GB available. I think maybe their may be something lurking in my registry.

Probably a stupid question but when you delete stuff do you let it sit in the recycle bin?

On that 500 gig drive, is all 500 gig in a single partition?

I generally empty my recycle bin. And the hard drive space is in a single partition.
