How can I get rid of infection?

Specs: Win7 64bit, Intel C2D/4gig
A chiropractor friend of mine gave me 2 Dell Dimension 4700 pc's to fix because they had some viruses/spyware. I could not run any antispyware programs while the dwrives were in the pc's. I took them out and put them in some external boxes, hooked them via USB to my machines and then ran Malwarebytes, Adaware, Spybot, Kaspersky, AVG, and Microsoft Security Essentials on both of them. When they were infected, the employess still used the machines for approximately 2 weeks until the error messages made the machines almost unusable. I have not used HiJack This yet. I think that I have one machine clean, but considering the 'residual damage', should my next step be either sfc /scannow or just a XP repair? I am losing patience as I am typing this. He has all of his business/customer data backed up hourly I think. I am just about to call him and ask if I can do a 'wipe and reload'.
Help. All opinions are greatly appreciated and respected.

I had also ran each program in 'Safe Mode' and did rid the machines of some infections but not all.
