I am working on an excel spreadsheet. How can I create a default "setting" when using a list from data validation?

What do you mean by a default "setting"?

Are you asking how to have the cell with the data validation start with the same value?

If so,

1 - Use a Named Range as your Source for the validation list (e.g. =MyValidationList)
2 - Select the cell and enter the following formula in the formula bar:

=INDEX(MyValidationList,3)

The data validation cell will display the third item from your list, but only until a list item is chosen and then the formula will be lost.

If you can't lose the formula, then I think that a macro will be required. We will need a lot more detail related to what you are trying to do if that is the case.
