Does turning harddrive off affect iTunes?

Specs: Windows 7, 4096
Okay, so this is the situation. For Christmas I am broadcasting an overnight Radioshow over Internetradio. I do this using Edcast and iTunes. This means I have to keep my computer on all night, which is not a problem. However, the power settings have the option "turn hard disk off after" (which is set at 20 minutes). If the harddisk is turned off, will the music still be played or will iTunes simply stop and resume when the harddisk is turned on again? Should I change "turn harddisk off" to never for that broadcast or doesn't it matter?

Yes and leave it that way unless your system is a laptop.
