Have a 64GB usb3 flash disk. Can I run an OS, e.g. Ubuntu, from that on my PC, if I have updated my bootsequence to recognise it ?

Yes. In fact, that's the standard way to run Linux on a Raspberry Pi or similar. It will be quite a bit slower than a hard disk.

Most distributions of Linux will have no difficulty running from an external drive. Running Windows this way is considerably more difficult and is officially unsupported.
