My new Android 5.0.2 phone has this setting:
Keep Wi-Fi on during sleep
- Always
- Only when plugged in
- Never (increases data usage)
Why does having Wi-Fi turned off during sleep increase data usage?

Is your data service enabled? If so then perhaps you have a few things running in the background which are regularly poling out. Location software for assorted apps.; sat nav/maps etc.? Presumably when wifi is enabled your phone uses that to connect to the internet etc. rather than data service; and makes its call outs that way?
When I'm home I disable my data service and use the house wifi entirely/exclusively for everything. When out and about I only have data enabled when I need it; using free or my phone service hotspots whenever possible.
The phone is not enabled yet. So far I'm just exploring what is
on it and adjusting settings, in hope of preventing it from doing
nasty things when I do connect it to the rest of the world. Some
of the first things I did were put it in Airplane mode and turn off
Location and Wi-Fi.
I guess you are saying that the "data usage" refers to data usage
via the cell phone ISP rather than data usage via the Wi-Fi ISP, as
if Wi-Fi data usage somehow doesn't count as "data usage".

Look at what Republic Wireless offers. It should help you understand how it works.
Using my o2 account as an example. Yes - if I disable my data feed from o2 (on my iPhone) and just use wifi (home or hotspot) any data I download etc. doesn't apply against my o2 data allowance. Similarly any app pollling out will do so viat house/hot spot wifi.
I think that what those settings mean is while wifi is off the mobile device will use mobile data
btk1w1 nailed it. When wifi is disabled it uses your data on your plan. I only use wifi, so mine is constantly turned on.
