Which port are you talking about.?
Back?
Front?
Hub?
port on an add-on card?

If it is a built in port on the motherboard usually located in the back of the computer or cabled to a front connection
Check in the BIOS to make sure all USB ports are enabled.

Add-on cards installed in a PCI slot have their own drivers and they may need to be re-installed.

Hubs can cause issues because of lack of adequate power source
