You should just be able to open a command prompt and type in 'defrag C:' and have Windows defrag it for you. If you cannot do it this way, or have Disk Defragmenter work, it is possible that you may have malware installed on your machine.
In that case, open your web browser, and go to http://www.microsoft.com/securityes... and install that immediately to begin cleaning your computer. Then, try defragging again.
