I keep getting a popup when closing Outlook Express - the one suggesting to compress files to save space. Recently, when accepting the operation, I'm getting another popup halfway thru the process saying it could not compress a folder because it is in use by another program - do I have some virus onboard that is operating from a folder in Outlook Express?

It depends really, on what Outlook is trying to compress. Are the files in particular being used by another program that you have open?
Go to Task Manager and shut down every program and application except for Outlook and then try again. If this doesn't work, it's likely that there's some other factor preventing you from compressing them.
I've never heard of a virus that only affects this kind of situation though. It's more likely a resource conflict issue than a virus I'd imagine.
