Disk management, make them a volume, not a disk, and span the volume across the free space of both hdds. I don't remember the specifics, but the process is in Windows help and on the microsoft site I think.

If you mean two physical drives, it can be done using a RAID configuration in "stripe" mode.
If you mean merging a partition/volume with another, you can use PartitionMagic.
