Doesn't that have to be turned on in the BIOS?

XP will recognise a Serial Mouse in Safe Mode by default providing the COM PORT has been enabled in BIOS.

Click on Control Panel/ System/ the Hardware tab and then on Device Manager. Hit the + sign next to “Mice and other Pointer Devices” and then select your mouse and open its properties. In the properties select Drivers and then choose Update Drivers. Look around and see if there is an option to “make this device work in all profiles”
Get a different mouse for that port. Turn the BIOS settings to ON not Auto (if that is possible).
