Assigning a specific drive letter??

Can I specifically assign what drive letter will go to my hard drive?? It is not the boot drive (i have C: which is win2000 and D: which is winXP) The drive i wanna change is a separate drive i use for file access on multiple systems, and i want to map it to a different drive letter that would not normally be assigned ( like S: drive) so that when accessing files that depend on finding files under S:\dir1\file the path is the same, since depending on the OS, it maps the hard drive either before or after the CD-Roms, so the letter changes. I know this is confusing, but hopefully someone understands what i am trying to say.
Also, I am going to clone all the contents of the drive to make a backup of it, i have norton systemworks 2002, does anyone know of a good program (better than what i have) for scanning the drive for errors/viruses and backing it up??
