I have a laptop with 2 separate printers installed (1 at work, 1 at home). The work printer (set as default) works fine even if I disconnect the USB and then plug it back in again the next day. However, when I go home and print and select home printer to print nothing happens. If I restart the laptop, suddenly it prints whatever I sent to it previously.
Any ideas on what is causing this? Presumably Vista has some bug in the printer spool service?

Does it make a difference if the (home) printer was set as the default printer? And have you tried an updated driver for the home printer?

I'll try but it shouldn't matter and the fact that the computer restarts without the default printer being changed means it's recognising the connection.
It's a hp laserjet 6l and was the latest driver installed by vista.

At least yours is still working, I retired my Laserjet 4L when it insisted on not working properly with Vista & HP has no plans to issue a legacy driver for it :-(
Oh & BTW, what are you using - Parallel to USB Cable?
At any rate, you may want to assign it to the USB virtual port 05 as suggested on this page.

It's assigned to USB002 virtual at the moment, not sure how to add a virtual port as ity seems it needs to be done during installation?
