Your model was first released 13-May-2008, only in the US and Canada.
It can't be any older than about 16 months.* Has anyone re-loaded Vista on this computer by using a regular Vista DVD?
Apparently your model is a desktop (tower) computer and has a built in memory card reader in the top bay.* Are you having a problem with a card plugged into that card reader, or another card reader that is plugged into a USB port?
If you are having a problem with a card reader that is plugged into a USB port.....
USB devices may not work correctly when they're connected to certain USB ports, e.g. ports in a hub or on the front of a desktop case.

Sometimes memory cards can be plugged in upside down. In that case they are not recognized, and they usually won't go inwards as far as they do when they're the right side up.
* Are you sure you're plugging it/them in with the right side up?* Did that same card or cards work fine in the same card reader before?
When you want to unplug a memory card, a USB connected card reader when it has a memory card in it, a memory card in a USB connected card reader, a USB flash drive, or a USB connected external drive while Windows is running, you are supposed to click on the Safely Remove Hardware icon in your taskbar on the lower right of your desktop screen, and choose to STOP accessing the device you want to unplug. That icon in Vista is a small mid green circle with a white checkmark on it - it may be hidden - you may need to click on the < at the left end of the icons in your taskbar.
(You can unplug any of those from a USB port when the computer is not running.)
* Have you been using the Safely Remove Hardware icon and choosing to STOP accessing the memory card while Windows is running when you want to unplug it?
* If the memory card came with a camera, or you bought one that was specifically meant to be used in a specific camera brand, what camera brand is it?
If that applies, or even if it doesn't
* have you re-formatted the card in Vista or by using something other than the camera's format feature to do that, and it was only after that you couldn't read it?Plug in the memory card, or one memory card at a time.
Go to Control Panel - Administrative Tools - Computer Management - Disk Management.
* Does the memory card show up as a drive on the right side there?
You may need to scroll down.* Does the memory card show up as a drive letter in My Computer or Windows Explorer?
* If it does, can you access it - can you read files you know have been placed on it?
