Since I have a whole bunch of programs which don't work on my new Vista PC, I made a separate HD partition with the intention to put Windows ME from my old PC on it. Can I just copy the entire C: drive from my old PC to the new partition, would that work?

no, all os' must be installed fresh so the registry can be created.

another thought, use vista program compatibilty feature. most older programs will run that way

Download from Microsoft's site VirtualPC 2007
by free and after installing a legal copy of Windows ME inside the defined virtual machine run your legacy programs under the guest ME environment.

Easier to do than say and high integration with Vista can be achieved (not to say no multi boot and separate partitions to manage).

all my drawers and boxes tomorrow to see if I can find the original CD's; I know they are around someplace...
