If your referring to the "The Microsoft Sound" (meaning you want to hear "The Microsoft Sound")Go to "Control Panel"> Double click "Sounds and Multimedia". Under "Sound and Events" heading find and highlight "Start Windows". Then under "Name" heading, use the fold down window to find and highlight "The Microsoft Sound". Then click "Apply" and "OK" at the bottom of screen.

If you can hear no sounds at all on your system unless you do the step by step startup or restart, you may need to reinstall or update your sound drivers.
If you have updated directX or other drivers without updating sound drivers this could mean you need to update (from the manufacturer of your pc or sound card, not Windows Update).
To reinstall sound drivers right click my computer icon>properties>device manager tab>click open the plus sign beside the entry for your sound device>highlight the entry you just exposed and click the remove button, reboot, the system should detect new hardware and reinstall the driver as Windows begins to load.
