Nope, well it's possible but use Format instead. It's alot easier. Remember you have to be in DOS-mode to do this, not a DOS-box.
Use: Format c: /q
This will delete everything(!!) on your HD.Use a bootdisk to get into DOS-mode!

To delete everything you have to format.
If your drive is c then go into dos with a boot disk select minumum boot or what ever, then at the dos prompt with the boot disk still in the a drive, type; format c: and hit the enter key. The computer will warn you that your data will be lost, hit y for yes and it will start formating the drive.
Good luck installing windows if you don't know what to do after.

POINT ONE
If you use FORMAT C: /Q you only wipe the MBR as it is a QUICK Format. Use FORMAT C: /U instead this is UNCONDITIONAL and will 100% Format your drive.
POINT TWO
Yes you must Format your Hard Drive from 'Real Mode' or better still from a Bootdisk. Windows must be shut down to carry out an effective 100% Format.

G'day Jack, if you have to ask that you should also ask "What do I save or check for before I nuke My System'[I'm sure Oprah did this one]. Win ME may not have some drivers for some devices so you should check to see if you have installation discs for all your devices first. Also check anything you may have created to save them to a floppy or whatever as well.
And as TechGUY more or less said, Windows will not commit suicide for anyone so forget formatting from Windows
And most of all , make sure you have a working Cd-rom Supported bootdisk plus your Win ME Cd-Key handy for reinstalling ME [not me, I'm not not getting the bullet yet]
As for Fdisk, if you are happy with one big drive or if you have multiple drives on the one HD and don't want to change them then don't even think about that one
and a Happy New Year/Century/Millenuim [Edition]/Epoch to you as well, only one hour to go down here in OZ

make a bootable dos floppy. copy format.com from your windows command directory onto it
boot from the floppy and then from the prompt type "format /y c:"
that'll do it for ya

Under windows?
1. open control panel
2. open "add /remove programs
3. open tab "startup disk"
Walla! 1 floppy and your done
