How many GPO's are being applied? This problem is generally indicative of too many GPO's to process. Possible solutions would be disabling portions of the GPO's that aren't being used, or if the server is over a slow link, enable "Group Policy Slow Link Detection. You may also consider removing links of unnecessary GPO's.

Also, run dcdiag to ensure your AD infratructure is healthy. See if there are any failures.

SOmeone recently added a bunch of new users to the intranet domain run on this server - could that be part of the problem?

What exactly is "this server"? Is it an app server? Domain Controller? File Server? What?

All of the above.
It runs a small office network (intranet)
Plus is the main server that runs the companies accounting package. Everyone was logging on with the same ID password, but they had someone setup 4 user accounts on the server - they hadn't rebooted in about 3 months, so it could be anything I guess. I went into SAFE mode and disabled all services and it did boot much faster - but it tells me little - no errors in the logs either..

Check to see if they are running APC Power Chute Business Edition version 6.xx. If so, they need to be updated (currently 7.04) which is available from APC's web site. You will need to create an account before you can log in. It is a 48 meg file.
