Slow file access

We have a HP server with smart array controller using raid 5 with 3 x scsi160 HD, I have a problem of accessing files from the server or client machine that sometimes the machines stalls for a few seconds when accessing network drives on the client or local drive on the server.
There are about 100-200 machines logged on sometimes. But I am sure not all uers will be coping or accessing file all at the same time.
The problem is if the server stalls for a few secs, all client machines on the network will also stall.
How can I get around this problem? is the bottom neck in the disc access?
When I do a disc defragment it will stall all client machines if they access the network drives also.
