I need professional answers only, am working on my 294.

You have three sites, but you have not modified the sites in AD, meaning that only the default first site exists. There are three, building 118, building 30, and building 1.

Building 30 is the wan link to both of the other buildings, which do not talk to each other.

The network looks something like this,

building 118
\
\
Building 30
\
\
Building 1

So if I where to specify that building 30 is on its own site and subnet. Would Building 118 use intersite replication to communicate with building one, or will it continue to send information every 5 minutes like they where not seperated?

It doesn't see to be an either/or situation. According to MS technet, buildings 1 and 118 would have to be added as sites to AD sites container in building 30 before any intersite replication or communication, can take place. The Knowledge Consistency Checker (KCC) runs by default every 15 minutes but can be changed.
