How do I require the username/password to be
entered every time one of my users connects to
my server running win serv 03

Consider self signed certificates and install on each system, change often for best security.

If using network shares secure by ntfs file permissions.

FTP is not secure even though a password is asked.

Consider webdave or other secure way to exchange credentials.

"Best Practices", Event viewer, host file, perfmon, antivirus, anti-spyware, Live CD's, backups, are in my top 10

tanasqui there is no file share password in server products. That is something that existed in peer to peer 9x networks.

You can purchase 3rd party software though.

Users should have to authenicate to your server [logon to the server via the network]. But this only happens once and doesn't happen again until they logoff and back on the network again.
