At work several of our machines have an icon that recently appeared that looks similar (but not identical) to the windows update icon. If you mouse over the icon the balloon says "Updates are ready for your computer. Click here to download these updates." Does anyone know what this is?

It's very likely to be a variant of a backdoor trojan. I just connected an unfirewalled PC to the 'net and was infected in around 5 mins. Look for a task named wininit.exe, running from WINNT\SYSTEM32, with an entry in HKEY_LOCAL_MACHINE\SOFTWARE\Microsoft\Windows\CurrentVersion\Run
It may have disabled DCOM, preventing network drives from being accessed - HKEY_LOCAL_MACHINE\SOFTWARE\Microsoft\Ole - check for EnableDCOM set to "N" and reset it to "Y"
