If a primary boot hard drive installed with Windows 2000 using NTFS file system. Can it be fomat in Dos mode?

No, DOS and Win9x can't even read NTFS. You can delete the partition if it's a primary one with Win98 fdisk, if it's a extended partition and there are NTFS and FAT logical drives on it, fdisk wouldn't even delete the partition. You can use partition magic or a small tool called efdisk to delete it anyway. Then you can put up a new partition with fdisk and format that one under DOS. But this partition isn't NTFS anymore, it's FAT16 or if you use Win98 fdisk and format and the partition is big enough it will be FAT32.
There are drivers for Win9x and also for DOS to read and write NTFS but only the read-only versions are free. Take a look at http://www.sysinternals.com/ to check what's possible with the right tool.
