Do you have an emergency reair disk?
In any case boot up from the Win 2000 CD and when it gets to the point of asking you if you want to re-install or repair, choose repair. If you have the emergency repair floppy, you'll be prompted to use it.
Among other things, an emergency repair disk can replace your boot.ini file with the one that's on the repair disk to get you back up and running.
I believe you can try to repair your existing installation, even if you do not have the emergency repair disk.
If you installed the Win 2000 Recovery Console when the computer was up and running, that would bail you out now too.
Give the repair a shot, even without the ERD if you don't have it.
Good Luck

As your problem happened while defragging, I'd suggest running chkdsk is a good idea. The boot.ini corruption may just be a filestore problem which chkdsk can sort (especially if you're using NTFS). You can run chkdsk from recovery console:-

http://support.microsoft.com/default.aspx?scid=kb;en-us;229716

I'd also run fixboot from here (repairs corrupted boot sector).

If this does no good, I'd suggest a repair reinstall (NOT a repair) - http://support.microsoft.com/default.aspx?scid=kb;en-us;292175&Product=win2000 (you need to start install as if for new installation - it should find your installation & offer to repair it. As you have SP4 CD you'll just lose windows updates since then (so visit windows updates!). It will leave your data, apps & settings intact.

Steve,
Just so I can learn something, you said "If this does no good, I'd suggest a repair reinstall (NOT a repair)"
I assume you mean not using the Emergency Repair disk? Am I right?
I was just curious what the reasoning behind that is.
