Virus or no virus?

Specs: Windows Vista
I have a basic question, if I ran a file that I know for a fact was a virus, but now norton 360 can't find it. Windows defender hasn't found a problem either. When i tried to run with the questionable file an error came up saying there was no file contents. I erased the files then did the scans so maybe thats why nothing was found. Is there another way to make sure it hasn't effected my files? Are there new viruses that can trick out windows defender, and norton 360? The only thing that may have changed on my computer is when i type a web adress in the adress bar of a open folder on my comp it doesn't open up internet explorer like it used to instead it gives an error saying the site can't be found. Browsing from an actual internet browser works fine though.
