There is a scenario where there is an application which uses Port 23. That port is open within the Organization. However that port is not open when connected over VPN. Is it possible to have 1 PC in the Organization network to run the application, then use RDP to conenct to that PC over VPN. Will this work ?

Yes, but tcp port 3389 must be open internal, and you get a ip address of your internal ip range. In this case, you'll act as an internal pc and can establish an RDP connection to that client pc.
