I want to stop my machine access to other persons.Using local administrator password others are trying to access my machine in my organization LAN. How can I stop this.

Do you have admin rights? Can you boot up into the default Administrator account? If so then simply change the password. If you are not the administrator then ask the administrator to change his password if you think other non-authorised staff are trying to access your machine. It probably isn't a good idea to try blocking the legitimate admin access to your machine.
