Our place of business runs a network using Windows Server 2003 and all computers run WindowsXP. I have two computers that "gum up" each day after 4:30 PM. The users cannot print to the network printer without a restart and files cannot be saved. The remaining 4 computers on the network don't have this problem.

With not knowing what is on the computers i would guess that a scheduled task runs at that same time everyday. It could be an inventory program like altiris or something like that. I would contact your IT person at your work as they would know what is running at that time.
