If it's a 2000 domain, then you can use a GPO linked to the domain. If it's an NT domain, then you'll have to use the policy editor to create a policy that you can then apply in the normal fashion.

Most corporate firewalls has option to prevent any download of file with extension: exe,scr,pif,vbs,... You should also set your mail server to remove such attachment.
