Am I correct to assume that in selecting a graphic card for a motherboard that has SLI capabilities I can either select a PCI-E if I know I will only use one graphic card or select an SLI type cards if I know I want to use two cards on the motherboard? Is that what the difference is between using SLI and PCI-E?

AGP or PCIe denote the type of slot/bus the card uses and has nothing to do with SLI capabilities. SLI is explained in the article, I recommend you read it.

I just recently bought an ASUS A8N-SLI Deluxe and to go with it, a GeForce 6600GT PCIe video card. Presently I'm only running the one video card but I might upgrade to dual in the future and it's nice to know the combination I bought (mobo and video card) is capable of it.
