Since Window isolates direct action to hardports I would recommend you try DOS first.

You need to be programming the UART (UART Universal Asynchronous Receiver/Transmitter) chip. As Wizard-Fred says, Windows wont allow you direct access to the hardware, you need to try DOS or perhaps some variety of Linux.
