Hopefully everything should go smoothly but you didn't tell us which board you're upgrading to or what processor you plan to install. Does it have an onboard sound, video, NIC, modem, etc.? Can't give info without info you know.

You shouldn’t have to reinstall your OS but you might have to reinstall all the drivers for your hardware. The new MOBO will have chipset drivers and you should install them first.
You can extract the drivers from your current computer by running this program:

When I swap motherboards I wipe out the current Device Manager entries so Windows can start with a clean slate. I do this by starting up in safe mode just before shutting down for the change. I start Regedit and delete the Hkey_Local_Machine/Enum entry. After the change Windows will pick up most (if not all) of the applicable drivers. You will need to install the new motherboard drivers and possibly one or two others.
