You should not have to enable it, it should just work.
Sometimes there are some settings in the BIOS that relate to AGP speed and/or timing and other things but I personally have never seen an enable or disable function.
Have you contacted ASUS?
Let me/us know how you made out as I would like to know how they treat you customer service wise. With so many wanting AMD systems and ASUS apparently being my most stable option with the Nforce chipset, it would be nice to know how helpful they are.

MY AGP knowledge is very limited, but my thought is that you plugged in a card that used a different voltage and blew out the AGP controller.

I have the same board. And I did a little research on it. It seems, that only AGP 4X and 8X cards are supported ( 1.5V). If the card is an older 3.3V it shouldnt work, and it can damage your motherboard. Other possible error reason is the power supply. (Some videocards need more power) a 300W supply is higly reccomended
