Uh, what? Do you mean in an SLI configuration?

You can run a PCI-Express system with just one video card. In fact, most systems available today will only support one card. There are a few (very few) on the market at the moment which support two cards in what is called SLI (forget what it stands for at the moment). Currently only NVidia cards are supported. I believe you can use differnt model cards. Even if that is true I would highly advise against it. The more differnt the cards are the more likely you are to encounter compatability issues.
