I have a USB Harddisk drive and require connection to it via a DOS boot disk, i have created a boot disk which detects both my USB CDROM DRIVE and Hard disk MSCDEX allocates the cdrom a drive letter but not the Hard disk, any ideas how to allocate this a drive letter? Any help would be appreciated.

You are probably referring to your local HD?

If you are referring to the USB HD here, you need drivers to detect the USB HD and map it to a drive letter. This assumes the USB HD has a FAT partition - not NTFS.
Check out x86' excellent link on various options. If your ext HD and computer USB port are USB 2.0, you could read on this link on HOWTO use USBASPI.SYS and DI1000DD.SYS drivers:
